Laemmli Buffer (2X) is an essential reagent in protein biochemistry, widely utilized for preparing samples for sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). Developed by U.K. Laemmli in 1970, this buffer remains the standard for denaturing proteins and ensuring accurate size-based separation. The buffer contains SDS for imparting a uniform negative charge to proteins, glycerol to increase sample density for proper well loading, and bromophenol blue as a tracking dye to monitor electrophoresis progress. Additionally, Tris-HCl maintains pH stability during sample denaturation and migration. When combined with reducing agents such as β-mercaptoethanol or DTT (added fresh by the user), Laemmli Buffer ensures complete disruption of disulfide bonds, yielding fully denatured protein samples suitable for reliable electrophoretic analysis.
